Major LC-MS Equipment

 

ThermoFisher LTQ XL linear ion trap
Two ThermoFisher LTQ XL linear ion trap mass spectrometers, one with electron transfer dissociation (ETD), are dedicated to routine in-gel protein identification. These instruments are each equipped with a Dionex nanoLC and New Objective nano-electrospray ionization source to enable the detection of low femtomole amounts of peptides in complex biological samples (e.g. whole cell lysates or fractionated biofluids). The ETD module provides an alternative method for fragmenting large multiply charged peptides and proteins. These instruments provide a low cost platform capable of analyzing more than 100 gel bands each week.

ThermoFisher LTQ XL linear ion trap with a multiplexed LC interface
ThermoFisher LTQ XL linear ion trap mass spectrometer is equipped with a multiplexed LC interface that supports automated 2D LC-MS/MS analyses. The PicoSlide / PicoChip interface was developed in collaboration with New Objective Inc. and facilitates high throughput multi-dimensional separations that improve the coverage and speed of shotgun proteomics experiments.

ThermoFisher triple stage quadrupole (Ultra and Quantiva)
Two ThermoFisher triple stage quadrupole mass spectrometers are dedicated to targeted proteomics experiments and equipped with a PicoChip nanoflow ionization sources. The Ultra is equipped with a Dionex nanoLC and used for projects that do not require ultra-high sensitivity. The Quantiva is currently the most sensitive of ThermoFisher triple quadrupole mass spectrometers and is equipped with the EasyLC nLC1000. This instrument is dedicated to the quantification of low abundant analytes (e.g. phosphopeptides).

ThermoFisher LTQ Orbitrap (Velos and XL)
Two ThermoFisher LTQ Orbitrap mass spectrometers are dedicated to high resolution proteomic profiling by Differential Mass Spectrometry. The types of samples that are supported by this instrument include plasma, CSF, and cell lysates. The instruments are equipped with Waters nanoAcquity nanoLC systems and PicoChip columns that provide highly reproducible nano-separations.

ThermoFisher LTQ Velos Orbitrap with ETD
One ThermoFisher LTQ Velos Orbitrap mass spectrometer is configured with ETD to support to high resolution MS/MS sequencing of large non-tryptic peptides or intact proteins. The instrument is equipped with a Waters nanoAcuity HPLC and a PicoChip nano-electrospray ionization source. The Velos LTQ Orbitrap is used to support Stable Isotope Labeling with Amino acids in Cell culture (SILAC) and Isobaric Tag for Relative and Absolute Quantitation (ITRAQ) proteomic profiling experiments.

Bruker microTOF ESI-TOF with HPLC
A Bruker microTOF ESI-TOF is equipped with an analytical HPLC and utilized as a general purpose LC-MS for the analysis of intact proteins and peptides.





ThermoFisher Q Exactive with Vanquish UHPLC
The QE is a high resolution/accurate mass hybrid quadrupole Orbitrap mass spectrometer. It is used primarily for our untargeted analysis for metabolomics, lipidomics and structural confirmation.





Sciex 6500 QTrap with Shimadzu Exion UHPLC
The 6500+ QTrap is primarily utilized for targeted analyses and in instances where MS3 is helpful in molecule identification. Additionally this instrument is more sensitive in negative ion mode compared to the 5000 triple quadrupole, which is essential for low level detection of bioactive lipids.


Sciex 5000 Triple Quadrupole with Shimadzu/CTC PAL HPLC
The 5000 triple quadrupole is utilized for targeted analyses of small molecules.



Major MALDI MS Equipment

 

Three Matrix Assisted Laser Desorption Ionization (MALDI) mass spectrometers are present in the BioMS Center. The Applied Biosystems 4800 Plus MALDI-TOF/TOF Analyzer and Voyager DE PRO MALDI-TOF instruments are used to support sample preparation, check digests, and quickly screen samples for specific peptides or proteins. In addition, these instruments are also used to determine the intact molecular weights of purified proteins and protein isoforms. A Bruker Daltronics Ultraflex II MALDI TOF/TOF MS is setup to provide basic MALDI imaging capabilities.
